Operation steps:

1. First take out the inner sterilization barrel, and then add an appropriate amount of water into the outer pot to make the water surface level with the triangular shelf.

2. Put back the sterilization barrel and load the articles to be sterilized. Be careful not to pack too tightly, so as not to hinder the steam circulation and affect the sterilization effect. The mouth end of the triangular flask and the test tube shall not be in contact with the barrel wall, so as to prevent the condensed water from wetting the paper at the mouth and penetrating into the cotton plug.

3. Cover and insert the exhaust hose on the cover into the exhaust slot of the inner sterilization barrel. Then tighten the two opposite bolts in a symmetrical way at the same time to make the tightness of the bolts consistent without air leakage.

4. Heat with electric furnace or gas, and open the exhaust valve to boil the water to make the water boil, so as to eliminate the cold air in the pot. After the cold air is completely discharged, close the exhaust valve and let the temperature in the pot rise gradually with the increase of steam pressure. When the pressure in the pot rises to the required pressure, control the heat source and maintain the pressure for the required time.

5. When the sterilization time is up, cut off the power supply or turn off the gas to make the temperature in the sterilization pot drop naturally. When the pressure on the pressure gauge drops to 0, open the exhaust valve, unscrew the bolt, open the cover and take out the sterilized articles. If the pressure does not drop to 0, open the exhaust valve, and the pressure in the pot will drop suddenly. In this way, the culture medium in the container will rush out of the flask mouth or test tube mouth due to the imbalance of internal and external pressure, resulting in contamination of the cotton stopper with the culture medium.

6. Put the sterilized medium into 37 ¡æ incubator for 24 hours. If there is no growth of miscellaneous bacteria, it can be used for standby.

matters needing attention:

Completely eliminate the air in the pot so that all the steam in the pot can be completely sterilized. There are several different ways of bleeding in high-pressure sterilization, but the purpose is to exhaust the air, make the temperature in the pot rise evenly, and ensure complete sterilization. Common methods are: close the vent valve. After power on, open the vent valve when the pressure rises to 0.05Mpa to release air. Close the vent valve after the pointer of the pressure gauge returns to zero. After closing the valve and re energizing, when the pressure gauge rises to 0.1 MPa, start timing and maintain the pressure at 0.1 ~ 0.15 MPa for 20 minutes.

When the pressure holding time is reached, the power supply can be cut off. When the pressure drops to 0.05Mpa, the steam can be released slowly. At this time, pay attention not to depressurize too fast, resulting in severe decompression boiling and overflow of liquid in the container. When the pressure drops to zero, open the cover, take out the medium and condense it on the platform. Do not deflate for a long time, resulting in changes in the composition of the medium, so that the medium cannot be inclined. Once placed for too long, the cover cannot be opened due to the negative pressure in the boiler. As long as the vent valve is opened, the atmospheric pressure enters and the internal and external pressure is balanced, the cover can be easily opened.

For articles that do not deteriorate after high-pressure sterilization, such as sterile water, cultivation medium and inoculation tools, the sterilization time can be prolonged or the pressure can be increased. However, the medium should strictly abide by the pressure holding time, not only to completely maintain the pressure, but also to prevent the deterioration of the components in the medium or reduce its effectiveness. In addition, the time should not be extended at will.
